长期从事海洋天然产物化学,化学生态学,以及生物合成研究,取得了一系列创新型成果。主要研究内容包括利用培养调控和基因组挖掘技术,从海洋特殊环境(冷泉、海绵、深海等)来源微生物中挖掘新颖天然产物,尤其是聚酮和环肽类化合物,评价其生物活性和化学生态学功能;综合运用基因组学、化学和生物化学等手段,解析新颖天然产物的生物合成途径与机理,开发海洋来源生物酶学资源。代表性工作有:(1)发展了基于培养调控策略挖掘海洋真菌天然产物化学结构和生物活性多样性,从一株散囊菌属真菌Eurotium sp. SCSIO F452中分离鉴定88个化合物,包括58个新化合物,且新化合物多以对映异构体形式存在,多个化合物显示出优良的抗肿瘤和抗氧化活性,研究成果发表于Organic Letters、The Journal of Organic Chemistry、Organic Chemistry Frontiers等杂志;(2)阐明了一类真菌来源苯二酚内酯的生物合成途径,利用组合生物合成手段在体内和体外组合表达两个后修饰基因与不同来源的苯二酚内酯合成酶,获得了十余个“非天然”天然产物,部分新化合物显示出优良的抗肿瘤和抑菌活性,研究成果发表于Journal of the American Chemical Society杂志;(3)发现了多个来源于海绵及其共附生细菌的环肽类化合物,揭示了部分化合物的生态学功能和生物合成机制。通过分子克隆、蛋白表达和纯化获得一个新颖的硫酯酶,利用体外反应验证了该酶催化吲哚氮原子成环的活性,相关研究成果发表于Organic Letters、The Journal of Organic Chemistry、Journal of Natural Products、ChemBioChem等杂志。
